I have a friend who work as a instructor in the school, he signed the 3 months contract of teaching profession. Now he is being appointed by our school director as a DTS (Dual Training System) organizer, Its a program from the TESDA pursuant to the R.A. 7686, and now he seeks for additional compensation for the position of being a coordinator.

Is he entitled for the additional compensation? If so, what will happen to the 3 months contract he signed?

No. Apparently, what was written in the contract was a broad definition, like "and other duties that was assigned to him", this was written in the last part of the paragraph of his duties.

he can request for additional compensation, but I would argue that if it is related to his initial tasks, it can still be assigned to him.
